Drink Containing Chlorophyll Extracts Improves Signs of Photoaging and Increases Type I Procollagen in Human Skin in Vivo

Abstract
Background: Although preliminary evidence from in vitro and animal studies suggests beneficial effects of chlorophyll on human skin due to its antioxidant and antimutagenic activities, no data exists regarding its effects on skin aging and acute cutaneous ultraviolet(UV) response.

Objective: Objective of the study was to determine whether dietary chlorophyll extract improves wrinkles and elasticity, and increases type I procollagen gene expression, and reduces UV-induced DNA damage and apoptosis in human skin.

Design: Healthy female subjects over the age of 45 were recruited and received 2 different doses (low-dose, 2 packs/d; high-dose, 6 packs/d) of chlorophyll extract supplementation for 90 days. Baseline status was used as control. At baseline and completion of study, facial wrinkles were measured using skin replicas, and skin elasticity by an in vivo suction skin elasticity meter. Skin samples were taken before and after chlorophyll extract intake to compare type I procollagen mRNA levels by real-time RT-PCR. Skin samples were also obtained 24 hours after 2 minimal erythema dose(MED) of UVB irradiation before and after chlorophyll extract intake to assess thymine dimer formation and keratinocytic apoptosis in response to UV.

Results: After chlorophyll extract intake, facial wrinkles and elasticity improved significantly(p< 0.05) in both groups. In photoprotected skin, type I procollagen mRNA levels were significantly increased in high-dose group, and procollagen immunostaining was increased in both groups after chlorophyll intake. UV-induced thymine dimer staining and UV-induced apoptosis of keratinocytes were substantially reduced in a dose-dependent manner.

Conclusion: Since chlorophyll extract significantly improves wrinkles and elasticity in photoaged human skin, with an increase in collagen production in photoprotected skin and decrease in UV-induced epidermal keratinocytic damage, it is strongly suggested that chlorophyll extract prevents and repairs cutaneous photoaging.
